Dunno about best, but my favorites would include:
Mireille and Kirika (Noir)
Sawa and Mikura (Kite and Mezzo, respectively)
the titular Gunslinger Girls
Kino (Kino's Journey -- not exactly a traditional gunslinger, but does sling a mean one when the situation calls for it)
Rally Vincent (Gunsmith Cats -- though more for the manga than the rather watered-down anime)

Wait, this is anime.
Vash the Stampede and Millions Knives from Trigun. Though I'd say that Knives is better in the long run simply because he doesn't have that taboo on killing but then again Vash is skilled because he knows which non-vital areas to aim at to keep the opponent alive.
They're both awesome.

Madlax. She doesn't even have to try. She shoots down a chopper without even looking. Throughout the entire series, 1 person knocks her gun out of her hand, 2 people can actually hit her with guns, and she only misses when she was firing at one particular person.

I also second Vash The Stampede... the best thing about his sharpshooting is that he aims perfectly so he doesnt kill anyone of his opponents...
Who else can do that?! :P
He's also the funniest gunman i've seen in anime. :thumbsup:

Anyone with collapsable rocket launchers, double-barrel blasters, a caster gun, and the infamous "revolving" bazzuka (and a ship with arms complete with a naked android in a box) definitly deserves to be on the list, and high on it. He is certainly not the best gunslinger, but in my opinion he is definitly the coolest, and he epitomizes the traditional gunslinger archetype better then most other examples. However one should expect something like that from Itoh...

But having the bigger gun helps, we've been shown that historically. If I were in a gunfight I'd definitly go Dr. Holiday and be the guy with the shotgun...

Not really. You said Vash is overrated, but I really like to know why. He really is one of the best gunslingers in all of anime so I don't see how you really can suggest that he is overrated.

I say it cause most people think he is the best and all, and vash aint even human more like a cyborg, and i would say Trigun is a medicore anime, that's IMO, so i overrate Vash, and i the first half of the series was good then they screwed it up near the mid to end of the series.

How is any of that relevant to whether or not Vash is a qualified gunslinger? By definition, a gunslinger (http://dictionary.reference.com/search?q=gunslinger) is "One who is armed with a gun, especially an outlaw."
Well, Vash is armed with a gun, so he meets the first criterion. He has a $60 billion dollar tag on his head, so he qualifies as an outlaw - the second criterion. The only question is whether he's the 'best' or not. To be considered 'qualified', he'd simply have to hit what he's aiming for. Vash generally does, way moreso than Spike Spiegel or Gene Starwind, anyway. So he's in the running, regardless of whether or not you find the series overrated.
The inability to separate one's personal feelings from a objective discussion of a character or series is, sad to say, a prime symptom of n00bism. You can cure n00bism by taking two teaspoons of Perspectival and calling your doctor in the morning. If you don't believe me, Ask Dr. Rin. :)

yeah, and alex rowe shot down an enormous starfish and two guilders who fell at the same time with a little .35 caliber pistol. it took him two seconds to do so and no one saw him coming.

Alucard because he is capable of effeciently handling two rediculously oversized hand cannons at once.
Rushuna Tendou from Grenadier can bounce rounds out of her boozum and into the revolver. Definatly a trick that wins first prize.

Sorry guys, but Vash loses big time. He was good at trick shooting, but when the time came to drop the hammer for real he always choked. Vash always balked when it counted.. and for those who can be honest about the series ending you certainly know what I mean.
Anywho, on to the list. In no particular order.
Emeraldas *The Entire Lejiverse* - Faced down a starfighter with a handgun.
Maya *Burn Up* - OMFG that's some firepower.
Chise *Saikano* - Does being a gun count?
Maria Tachibana *Sakura Taisen* - Being able to shoot bullets has to count for something.
Rally Vincent *Gunsmith Cats* - I have to learn that foot reload trick.

She's like Kenshin only with a gun. She often moves behind or beside people instantly, like Kenshin, or even Alucard. She catches bullets 6 at a time in the chamber of her gun and dodges rounds all throughout the show.
Plus she's got an incredible body. I'd hit it... gently. :wub:

Tendou Rushuna from Grenedier if only for her unique reloading method. Outside of that Rally Vincent is my favorite, with Triella from GSG a close 2nd and third is Sawa from Kite

Train Heartnet (Black Cat)- for a teenage guy is an excellent, deadly marksman who never has failed his duty as a number of Chronos and rarely misses his target.
Levy (Black Lagoon)- is great for her superb ambidextrous use of guns. There is a reason why she is called Two-Hands.
Henrietta, Triela and Rico (Gunslinger Girl)- they aren't called gunslinger girls for nothing. They use guns because they enjoy using them. And they are not only efficient but they are also ruthless and almost coldblooded gunslingers, ruthless enough to kill terrorists without remorse.
Maria Tachibana (Sakura wars)- is even able to deflect a bullet by shooting it with an another bullet
Rushuna (Grenadier)- is a golden-haired Senshi--a gunslinger who travels the land with one purpose: to make the world a peaceful place. She is even a better gunslinger than Maria Tachibana. Rushuna not only can shoot her bullets against the bullets of her opponents but also when firing a single bullet against one of her enemy's bullets, the fragments of her fired bullet in collision with the single enemy bullet shattered all of the other bullets
